反义技术分子式：CAS号：性质：根据碱基互补原理，用人工合成的或生物体中自然存在的寡核苷酸片段(即反义寡核苷酸antisense oligo nucleotide，指反义DNA和反义RNA)或其化学修饰物，结合于目标基因或mRNA上的特定序列，抑制或封闭基因表达的技术。反义作用有专一性和非专一性之分。专一性反义作用主要发生在五个层次：(1)抑制DNA的转录；(2)抑制前体mRNA的拼接；(3)抑制mRNA的戴帽及poly A尾巴的连接；(4)抑制翻译的启动；(5)阻遏核糖体在mRNA上的移动。在病毒里存在非专一性反义作用，其本质是诱导病毒的RNase H非专一性降解病毒mRNA，以抑制翻译的进行。在针对转录过程的专一性反义抑制中，反义寡聚核苷酸与双链DNA的某一段特异性结合，形成三螺旋结构。无细胞提取液等体外实验表明：三股螺旋结构能抑制DNA的复制，阻断转录因子的结合，并阻断转录的启动。依据三股螺旋结构设计反义寡核苷酸有四条原则：(1)最佳长度为20～40bp；(2)结合区段富含嘌呤碱基的那条链的嘌呤含量应在65%以上，以确保三股螺旋的结构的稳定性；(3)遵循G与G、C形成GGC，T与A、T形成TAT的原则；(4)结合区段中，若富含嘌呤的那条链中嘌呤含量过高，构建的反义寡核苷最好能与该链的反向平行。针对翻译过程的反义调控包括：干扰mRNA与核糖体的结合，干扰mRNA的构型、剪切拼接、戴帽与poly A尾的连接、激活RNase H降解mRNA等。这一技术从反向遗传学的角度认识结构基因的功能和基因表达的调控，从而为分子遗传学分析、人类疾病的防治以及动植物遗传育种等提供了崭新的手段。
